It means that since you're looking for the job and you have gone through the trouble of submitting a resume, you will follow it up with a phone call.

This is an indication that you pay attention to detail and follow up all projects to ensure they are complete.

Indicate that you will call them during business hours in a few days to confirm that they have received your resume and get their feedback on the job and are prepared to answer questions at any time.

You should always call, but don't tell them that in a letter.

The cover letter should end with "I look forward to your response" or "I look forward to meeting with you to discuss the mutual benefit of our working relationship" - this lets them know you will await their response, but only wait a set period of time. If you hand in a resume/cover letter, ask the person's name who is taking it, then you should give them two days to look it over, then call. Use a professional tone, asking to speak to the person who does the hiring. Say something like, "I left my resume on with and I was hoping you had a chance to look it over; I'd be happy to answer any further questions you may have..."

If you mail it in, wait for four or five days to allow for delivery, then do the same thing. "I mailed my response on to ...)

It's true the squeeky wheel get the oil, but don't seem so pushy that you tell them you are calling them.

You should do the calling.

Just indicate that you would like to call them a few days after they receive your resume.

You do the calling. They should have used a comma after the word ---letter. It is not required (the comma), but it would have made it clearer.
