Are you constantly updating programs, are you on Vista. If you mean the computer desktop refreshes it usually happens when a major process ends or when the computer's memory get to low. I don't think it is anything to worry about your computer may just be recvieving updates a lot, check your settings.

as stated, this could be thousands of things, without knowing what you have on your computer, this is not easily answered. however, 2 thigs could help you here. Task Manager and MSCONFIG.
When looking at your desktop, press Ctrl+Alt+Del one time together to bring up the Task Manager. There are 5 tabs at the top of this window, but only 2 really concern this issue. viewing the Task Manager changes nothing on your computer but ending things can produce undesireable results.
Applications:
look at this tab to see if any programs are openly running, if you are sure that a program is running in this window that shouldnt be, highlight it and click "end task" at the bottom.
Processes:
This window will show you all that Windows is running (as well as the files for what You are running) this list can be large. click first the CPU column head to arrange the list by CPU useage. usually if your Hard Disk is doing alot, so is your CPU. when you find something working overtime (except System Idle Process) then what I do is look up the image name (file name) on the net and find out what is using that file, and go to the program using it to make changes. many time you cant tell what the program is by the name, but be very careful to spell the name exactly as it appears when searching, many viri use similar names to real file names. let the net tell you what your specific problem is and how to cure it..
MSCONFIG
search your Windos folder for this application. when you have it up, you might be able to stop bad programs from starting automatically. ONLY change things in the STARTUP tab. the other tabs are for those people who reeealy know what they are looking for..
In the startup tab, you may check or uncheck items which you want to start on your computer every time it runs. some items may be necessary for other programs, and some may be completely unnecessary. a restart is always wished if a change is made, it is also a good idea. you will get a warning window when you return, but this is just a heads-up that you changed something..
Good Luck!!

Do you mean the computer is clicking, like the 'refreshing' sound that internet explorer makes?
It could be any number of things, and I cant really tell without hearing it, but the main thing I think of is the hard drive spinning constantly. It could be that something is constantly scanning the hard drive, such as an antivirus program or some sort of malware. Check your antivirus and if its not that, download something like adaware or spybot search and destroy to look for malware.
