If you work in an environment where there are close family members, for instance husband and wife (and the wife just happens to be boss) and her husband happens to be your line manager, what instances of conflict of interest would there be?

You may find it difficult to complain to your boss about your line manager as they are married but that's the only thing I can think of. Did you have any problems with them yet? If not then don't dwell on something that may never happen. Presumably they are professional people who want their business to succeed.

If you are employed for your skills it doesn't matter to you at all.
If you offer a service you can only be loyal to your line manager and if you think he is a wimp find another boss.
You can't please two people and if his wife starts dealing with you over his head tell her.
No, or make me your husband's boss and then I'll listen.
Make yourself indispensable and give them two fingers.
Mind you, she might have other things in mind, if so get out now.

It might be difficult to go over your managers head if you have a problem.
