Not its not painful. It can be done 2 ways, first is by a mid stream urine sample (MSU), and the other is by a swab. In one hospital where I worked, we would let the woman do her own swab. The swab looks like a long cotton bud, and you just insert it into your vagina. If there is a reason to perform a vaginal examination on a woman after 34 weeks, then your Midwife or Dr will do the swab. However, there should be no reason to perform a vaginal examination unless you have had contractions before 38 weeks, or you are over due and they will asses you to see what they need to do to get you into labour. Many vaginal examinations during pregnancy are unnecessary, you should always ask for them to explain why you need to have an examination, if you are not in labour, and have not had any problems.
Some Dr's will insert the swab into the vagina, and then swab down over the perineum, and then down into the anus, as that gives the best indicator if there is GBS present (Group B Strep). Many hospitals are no longer testing for GBS, as it changes over time, so you may have a positive result at 38 weeks, and a negative result at 40 weeks.

Tested negative every time, it is simple they use a Q-tip like swab (like when you have a pap smear), but they don't insert a speculum to do it. So it is not uncomfortable at all. You will just lie back like a pap exam and they just barely put the swab in (but be warned they will more than likely swab your anus quickly also) but it's over in seconds. This is usually done around the 35th week of pregnancy.

Not painful at all... a little uncomfortable.. the dr will probably do it between 34-36 weeks. It's just a test swab, not like a full pap. Even if you test positive, don't worry. I tested positive with 2 of my 4 babies and they just give you antibiotics while you're in labor, both of my kids were fine. :)

I don't remember being tested for Strep B. I tested positive during delivery and my son was born with Strep B and Pneumonia. I was told that I delivered to fast for the medication in the IV to reach my son.

It is just like a pap, had mine done today.
around 36 weeks but not before because they want to know if it is present at birth. It is harmless to the mother but dangerous for baby.
I'm 37 weeks

It doesn't hurt at all. It's just a small cotton swab they wipe around in you to collect a sample. No big deal at all.
I remember they did mine pretty close to full term. maybe around 36 weeks if my memory serves.
