The movie 300 is about the Battle of Thermopylae, in which 300 Spartans held off an attacking Persian army long enough for the other Greek states to prepare to defend Greece against the Persians. In the end the Spartans were annihilated but the rest of the Greek states were successfully defended.

The 300 Spartans were 300 soldiers from the Greek city-state of Sparta. Led by Leonidas, one of the two Kings of Sparta they were sent to stop the invading army of the Persian Emperor Xerxes. Xerxes was invading, partly at least to avenge his father Darius' previous defeat at the battle of Marathon and partly because the Persian empire was the biggest and greatest empire of it's day and you don't get to be an empire without expanding your borders (often aggressively)
The vastly outnumbered Spartans met the Persian army (rumoured to be numbering anywhere from 100,000 to over a million) at the pass of Thermopylae. The battle lasted for 3 days before the Spartan force was out-manouvered and slaughtered.

For the latest why don't you see the movie "300" on the subject. However, 300 were Spartan warriors against the might of Persian army who fought valiantly as in the Greek mythology as there have been regular battles between Greeks and Persians for many centuries.

Sparta is a city in the South Peloponese, Greece and an important city -state in ancient times, the greatest opposant of Athens. The 300 Spartans, led by general (not king) Leonidas, along with a 700 Plateans (who are very unfairly usually forgotten) went to the narrow pass of Thermopylae, knowing they would all die without any doubt, in order to hold the persian army wich was a 1000 times bigger, for as long as possible, giving time to the rest of the greeks to prepare their defense together against one of the persian empire attempts to put a foot on Europe.

Spartans were an ancient civilisation who's main aim was to be the best warriors in the world. The 300 you are referring to are 300 Spartans who tried to stop an army of 1 million invading Persians.
