Nope. The only thing that makes a person gay is if they are sexually attracted to the same sex and want to be physically intimate with them.
You'll find that there are gay men who don't like anal sex (look up g0y, for instance), and tons of straight men who enjoy anal stimulation, provided by their wives and girlfriends by means of fingers/fists, toys like dildos, or strap-ons.
Enjoying a part of your body doesn't make you gay. Wanting someone of the same sex to help you enjoy that body part does.

Probably if you do it with another boy.
Or maybe some people just try it out to see what it feels like.
Then if they like it, they naturely want to do that.
