Dodge Man is right (as usual) I know when I had the Flex plate and Rear Main replaced in my Suburban, we were able to push the transmission back far enough to get the room we needed to work, but you may not have that kind of room to work with. So my guess is, yes, you will have to pull the tranny to do this, I suggest the tranny because it it marginaly easier in most cases.
And not to be rude, but if you are asking how to replace it, you probably dont want to start messing around with pulling the tranny, you could run into some big problems that you may not know how to deal with, you will want an ASE certified mechaninc to perform the work.
Also consider if it is needed, its ALOT of labor charge for a very cheap part. if you find a good shop, proabably get it for under $350. just letting you know, so your not shocked if they give you a pretty steep price.
For help finding a good mechanincs shop, if you odnt already have one you use. Look for an ASE certifiction to be displayed, and aslso check the Better Business Bureau for a list of trusted shops in your area.

You will have to pull the transmission remove the flex plate, remember when putting the seal in put oil on the outside of the seal for easy installation so you won't damage the seal.

before replacing the rear seal ,replace the PCV valve (in top of the valves cover) in top of the engine.
and run the engines a few days, to see if the leakage has stop.

i own a repair shop and you either have to pull the engine out, or the transmission the choice is yours on this one,but one of them does have to come out to get this one in,good luck with it,i hope this helps.
