Something that will almost always get you the job

First off, you need to think about your availability...and you have to be completely honest. Managers like to hire people who don't have a lot of restrictions on their availability, but they'd rather you were honest with them. A lot of times, you can find lenient employers who can work around your schedule.
Second, you need to think about what skills you've got. The best things to put on your app would be great people skills, leadership skills if you have them, and anything else that will be helpful in the job you are looking into. Think about everything your job will require and which areas you have experience in. The more you include, the better your chances.

These are the best things to include:
Great people skills, leadership skills, follow directions well, hard worker, work well with others, friendly, etc.

Most applications don't leave you a whole lot of room to include everything, so having a resume definitely helps. However, the resume will mean nothing if the application doesn't look right. So make sure you include as much as you can on it and be honest. It's much better to be rejected for a job than to have to search even longer and harder for the next job if this one fires you because you didn't stand up to what your application stated.
Also, if you were fired from a previous job, state that you were Involuntarily Terminated (looks better on the app that just "fired"), and state that you will be happy to explain. Then when you get your interview, they may or may not ask you about it. If they do, just give them a straight-forward answer (they may check into it). Tell them exactly what happened and try to keep from shifting blame, but at the same time don't let them think that it was all your fault or that you're incompetent. Trust me, they'll appreciate your honesty and will likely see you as a trustworthy, reliable employee.

Good luck!

Umm your name.

Sorry, but no one thing will get you the job if you are not more qualified than the next person walking in the door.
