Fossil fuel depletion. It will lead to starvation and war, possibly killing billions of people, during the late 21st century.

However, a better way to give the same answer is "population overshoot" (relative to the resource base). The reason that's a better summary of the biggest threat to humanity is that fossil fuel depletion might lead to a less grievous calamity than would result from having an infinite terrestrial supply of fossil fuels.

Unlimited energy would lead to a choking of the world with heat and pollutants, which would sooner or later render the entire Earth uninhabitable. The petroleum deposits aren't too small. The human species has become too big. The world is now an overcrowded lifeboat, and the time for triage has come.

So the biggest threat to humanity, basically, is that there are too many humans. Those who answered "HUMANS" to your question are right, though perhaps not for the reason they thought.

Disease is historically the biggest threat to humanity, and is gaining new importance as our ability to fight disease is waning. Antibiotic treatments are proving less and less effective, and newer antibiotics are extremely expensive and often nearly toxic in themselves.

Famine will also prove a major threat in the future, as it was in the past. Our elaborate production/distribution networks are very vulnerable to disruption, and any disruption is likely to prove catastrophic within a few days.

A single hydrogen bomb could flatten the entire New York metropolitan area. An exchange of hydrogen bombs would throw so much earth up into the atmosphere that before it came down as fallout, it would block the sun, causing nuclear winter. All the green plants would die and everyone on earth would either starve or freeze to death.
