I configured a file server in ubuntu 6.10 with samba. I can access the foulders in windows with different privilege, etc; everything works fine. The next day I can't access the server. I reload samba and nothing.

Is not the first time I got this problem. I change the computer thinking the problems was the (old) machine but in the faster computer got the same situation. Whats wrong or how I can fixed?, thanks!

Check the firewall on the windows machines.
Then see if you can ping the samaba IP. If you can't you need to trace out the correct location for it.
I hope you gave your sever a "fixed" address within the LAN, if not, do so. Give it one outside the DHCP range of your router but still within the network addresses range.
