Well if Mario and Luigi got a invincibility star then that would shift things in their favor, but I don't think Spidey or Iron Man would let that happen. Spidey's senses would go off when the star came out. But then again Mario and Luigi both have 3 lives a piece to figure out a strategy.

Ok, ok, stop. Mario and Luigi rule, this is true. But you're talking fighting superheroes here! That's not even fair. Videogames and Superheroes are 2 totally different areas. Except superhero games, but they're still fighting villains above Mario and Luigi's level. Granted, if they got an invincibility star, they could win, but I don't think that'll happen. Sorry.

Hahah. Good question. :)
IronMan and SpiderMan have super powers. Super agility, laser-shooting costume, web-throwing.
Mario and Luigi can jump out of the map ( that means friggin high I'd say ), transform into a racoon, shoot fireballs, eat only ONE mushroom and grow bigger ( bigger not FATTER ), get a flashy star and no matter what the hell is in their way, they won't God damn die.
It's a very hard battle. OH ! IronMan has kicked Mario, but Luigi gets over his head. OH THE IRONy. :))
And the winner iiiiiiiiiiisssssss .. LUUUUUUUUUUIGIIIIIIII AND MAAAAAAAARIOOOOOOOOOOOO.

Mario and Luigi... they'd just get yoshi to eat the others and make eggs out of them...
or they could throe fireballs... or they could run over them with a cart...
or eat a mushroom and then just step on them....
simple...
