Queen Elizabeth is Prince Charles mother not his sister.
When Elizabeth's father, King George VI, died he had two daughters and no sons.
Therefore, the elder daughter, Princess Elizabeth, became Queen Elizabeth the second.
There was an earlier Queen known as Elizabeth and that's why the present Queen is Elizabeth the second.

By tradition the first-born male to the reigning monarch is first in line to the throne followed by the second born male and so on. If the line runs out of male heirs then it falls the first born female heir.
Also, by tradition, an heir to the throne cannot marry a divorcee which was, in part, why Edward VIII abdicated (the other part being that Wallace Simpson was a commoner).
After Prince Harry, the next in line is Prince Andrew.
There have indeed been two King Charles'
Charles I was the son of James I and was executed on the orders of Oliver Cromwell in 1649, beginning the Interregnum.
Charles II was the son of Charles I and spent a period of exile in France until 1660 and the restoration.
Many people foolishly believe that The Queen Mother was Elizabeth I. However, The Queen Mother (Elizabeth Bowes-Lyon) was Queen Consort of George VI, the actual Elizabeth I (Elizabeth Tudor) was Queen Regnant from 1558 until 1603.

There WASN'T a male heir when Elizabeth took the throne. Her Uncle Edward had abdicated, in order to marry Wallace Simpson.
And then, it goes down to Elizabeth to have her heirs take the throne upon her death. That would be Charles as first born.
I thought everybody knew that!

King George VI (Elizabeth's dad) didn't have any sons. In the British line of succession, boys only have precedence within a family - i.e., a son will ascend before a daughter, but the daughter of a king still has precedence over more distant relatives (such as brothers or nephews of a king).

The throne is still occupied by the queen,when she dies or steps down,then and only then can prince Charles sit on the throne, cheerio old chap
