My MB is an ASUS A8V, and I want to add more hard drives, but I've filled all my SATA slots -- and I'd rather go SATA than IDE. So I bought a Rosewill RC-211, which is apparently a PCI-E card. Can I use it on that motherboard?

I'm not sure about the A8V as I don't have the documentation with me. How ever PCI-e will not work on regular PCI. They are two different size ports. There's re several types of PCI-e and they are mustily used for video cards. Hope this helps.Will.

That board has PCI-E but is the appropriate slot used by a graphics card?

No, pci-e wont fit or work, you should return it and get a regular pci sata controller for your motherboard
