i dont think you can set the passwork for only one folder
but this is what i normally do

right click of the folder and you will see "hidden" put a click next to it
or then you will see the advanced
you will see encrypt content to secure data

and then when you see sharing
you will see "make this folder private" and click on that,
it will probably make it to be private and only you can get to it

or if not, then you can create your user accounts and set up the password so then only you can login to the computer
unless you turn on the guest user if not then only you or admin can get into your computer

Create another user account by doing this:

1) Right Click on My Computer and select Properties
2) Expand the Users and Groups folder
3) Right-Click on users and select New User

Go to the folder in question and remove all rights assigned and reassign rights only to the user you just created.
