too much running in the background?
long time since it was 'cleaned out'?
norton?
game playing?
virtual memory filling up? (getting low on disk space)
overheating?
many causes to this, first thing i would do is to stop all programmes from starting on boot up except anti virus/firewalls.
(search for msconfig go to the end tab and uncheck everything except the anti virus/firewall)
some games and utils will keep using virtual memory rather than flushing it out, as a restart solves the problem this will flush the virtual memory....could also be getting low on disk space, by default, windows takes a certain % (10% i think) for virtual memory usage.
a good clean out or maybe a full format and reinstall can do wonders for your comp and on a 6 year old comp that might never have had a format this will give your old speed back.
every time you install and uninstall a prog, some rubbish always remains in the registry, as windows scans the registry constant, this can slow it down....but i am thinking more along virtual memory probs here, remove anything you dont need to free up disk space and see how that goes.
...but would suggest a full format and reinstall if just for the pure good it will do your comp

If the batteries in your mouse are fine, check your temporary files by running Ccleaner. Use the Cleaner tool when you boot, then again 2 hours later. If theres a lot of files (megabytes) the second time, theres a problem!
Run an anti virus and anti spyware scan in safe mode.

And use the Tools - Startup list to cut down the programs that load on startup, leave Windows and your security software, but stop everything like your office suite. The programs will still be available from the Start menu.

Hmmm it may be overheating (either CPU or graphics card)? check that your cpu fan is coming on? you might wonna give it a clean and get dust out.
Also check the temp of you computer in bios and watch to see how much it rises (if its a desktop computer over 60*c aint good). Find out what cpu you have and then type on the net 'max temp of _ _ _ cpu'
This is a problem i had with a previous computer and found that it was overheating :)
