To prevent & remove spyware, use "Spybot Search and Destroy" (http://www.spybot.info/). This is free and very effective.

To stop pop-ups, you can use a browser like Firefox instead of Internet Explorer (although recent versions include pop-up blockers - it may be spyware causing the pop-ups)

load several spyware detection programs, they seem to work better if you have several, like spyware and windows defender. then use them on a regular basis, turn popups off in your browser, don't open mail or attachments that you do not know about, keep a firewall either hard or soft running and don't download music, movies from the p2p sites since many of them are infected.

use mozilla firefox as your web browser to limit the amount of bugs that make it onto your pc through software holes:
http://www.mozilla.com/en-US/products/download.html?product=firefox-2.0.0.2&os=win&lang=en-GB

and then do regular spyware scans to catch any that get through:
