Is it enough to prepare 20 replicates of each sample to get agood data? Please some idea just to give me confidence in my figures.

Unfortunately it not that cut and dry. The basic answer is 3. However this compeltely depends on the variation you will be experiencing. It requires some advanced statistics to figure out appropiate sample size.

if you have good technique for streaking for isolation, you only need one.
