I think our true hearts desire changes throughout life. Before I was a parent I would say my true hearts desire was to be a mother. Now that I am a mother I'd say my true hearts desire is to give my daughter the best life possible and for her to be happy.

To find your true hearts desire you need to sit alone and picture that you have all the money in the world in front of you and you can do anything, what would you do, who do you see in the picture- family, friend, pets, kids car, house travel, where are you. Write it down.
Now what is your hearts desire?
Find photos that relate to your desires put them on a board and look at them everyday, picture you self living in the home, car, what ever you are doing. see your self their and work towards that.
Watch the movie "The Secret"
Follow that path.

I beleive you and everybody knows this, but they often ignor it and choose a road that is easier, more convenient or just plane easier to see. Everyone has an interest that they are passionate about and that is the direction where your heart's desire lays

Honestly, I don't think you'll ever find the answer to that question because I believe we will always be searching for it. Perhaps the best you'll find is peace within yourself and when you're old, perhaps you can reflect on your life and say, "Everything I wanted to do, I did. No 'should've, would've, or could've".
Good luck though. I doubt anyone can really say they have no regrets or doesn't wish for a "do-over".
