I had 3 shot of tequila and beer 16 hours before taking a pre-screen drug test and I am concerned that I will fail the test and lose the job offer.

Usually they don't include alcohol. If all you did was drink you should be okay.

If a pilot can fly a plane with a 12 hour rule, you should be fine with 16 hours and really not that much to drink, unless you are four feet and six inches tall weighing 60 lbs or less, then you may be totally out of luck. Is this a religious job? Does it involve heavy machinery? Alcohol isn't illegal, so what if you had 4 drinks last night? I would be more concerned with that joint you smoked at the Phish show last weekend than enjoying your burritos and tequila last night.

Don't sweat it, if they give you problems tell them you used mouthwash because you wanted to make a good impression and ask if you can get retested, if you still want to work for a company that doesn't allow you enjoy your right to a couple cocktails on your own time.

Alcohol is a drug.
The pre-screening process only screens for drugs that the potential employer requests, as each screening is specific for a certain drug and each screening costs money.
If the employer specifically requests a screening for alcohol, they'll look for alcohol.

However, in a normal sized human, one dose of alcohol (1 shot of spirits, 1 glass of wine or beer) will be absorbed by the body in about an hour. Four doses 16 hours before screening (assuming you urinated a few times before the screening) will show as well under the legal limit, if it shows at all.

Your potential employer still has the right not to employ you if there are any traces.

You should be fine , your body should have cleared the alcohol out of your system in that time as the quantity you drank wasn't much. Bit silly though if you knew you had the test coming up so soon...!
