I'm new to stamp collecting and want to know whether stamps that are canceled (i.e. stamps that are marked from being in transit from the postal carrier) lose their value because they're marked? Or is collecting mainly reserved to unused stamps?

Both mint and cancelled stamps are equally collectable, up to a collector's choice and budget. Some collectors believe that a stamp is not a stamp until it has seen postal duty and cancelled, while others believe that cancellations obliterate the design. Generally, mint stamps are worth more than cancelled stamps, but there are exceptions where some stamps in superb used condition with rare or special cancellations from a certain location or of a particular event being more sought after than mint. I suggest that you acquire some stamp catalogs like Scotts or Stanley Gibbons to help you organize, and for the identification and valuation of stamps.

Certain cancelations at a specific historical times and places have a special value and will be considered more valuble than the base stamp itself!

When stamps are franked(cancelled)they are virtually worthless.They must not only be unused but in pristine condition.
