Usually the sea level is taken as a reference to express the pressure value of air.So the layer in touch with the sea surface can be taken as the of layer where the air pressure is greatest. If you want a specific place, the layer of air adjacent to the bottom of the deepest valley or the deepest part of the land in the world can be cosidered as the layer having the greatest air pressure.

Air pressure is the accumulation of 'atmosphere' being pulled down towards earth by gravity. When you leave our atmosphere (outer space) there is no atmosphere, and therefore little to no pressure. This is in part why parts of the space shuttle can be as thin as a piece of tinfoil.

Therefore, the highest pressures are right at the surface of the earth, where us humans and animals are. The huge column of atmosphere above us gets heavier and heavier as you get closer to the surface of earth because more and more atmosphere is 'piling up' on us as you get closer to the earth.

Alternatively you can think of it as this... when you climb a mountain you're higher in elevation and the pressure there is smaller than it is at sea level. This is because you're farther away from the core of the earth where the center of gravity is.
