One of the original federal routes, US 66 was established on November 11, 1926, though signs did not go up until the following year.[2] It originally ran from Chicago, Illinois through Missouri, Kansas, Oklahoma, Texas, New Mexico, Arizona and California before ending at Los Angeles for a total of 2,448 miles[3] (3,939 km).

"(Get Your Kicks On) Route 66" is an American popular song, composed in 1946 by Bobby Troup and first recorded that same year by Nat King Cole. The 2006 Disney/Pixar film Cars portrays the legendary road, Route 66 as a spectacle of the United States. It is remade by John Mayer. The origional singer I believe was Bobby Troup. The big deal with it is it is very historical highway that runs threw the center of the U.S. east to west. The movie Cars is based on true facts and characters of Route 66.

RT. 66, two lane highway, was once the main route East and West, before the Interstate system was built.. Some sections of the old 66 route still exist and can be traveled through Missouri.
I think it was a remake by Mayer.

As for the big deal, I think It's all about freedom. The combination of american car culture and the wide open west.
Route 66 was the major route from from Chicago to LA before the Interstate Highways were built.
