What was the cellular mutation that caused this. Is it known yet?

What reptiles? If you are referring to dinasaurs, they are not reptiles but a category unto themselves.

As for the dinos, they are believed to be warmblooded. The book The Dinosaur Heresies by Bob Bakker is worth reading. It will explain your question to you. Also, try going to a museum and you will learn much there.

Monotremes (egg laying mammals) have much lower temperatures than placental mammals.
Marsupials have a slightly lower temperature.

It appears to have developed in stages.

There are an absolute myriad of mutations which resulted in this, not just one. I doubt it will ever be known.
