When Star Trek creator Gene Roddenberry was asked this question, he replied that there was no significance to the letters and numbers comprising the registry of the U.S.S. Enterprise. At the time Star Trek first aired, airplanes commonly had "NC" on them, and adding the extra "C" updated the look. USS stands for United Star Ship.

NCC means nothing else than being part of the standard registration of star ships, just like NX is for protypes or experimental vessels.
USS stands for United States Ship, in Star Trek you also have HMS ships, and those 2 have real world counteparts. HMS ships are British ships, their is some Star Trek ships that uses HMS (his/her majesty's ship) before their ship names, like HMS New Zealand. Since we all know that Earth in the 22nd century is United, those désignations of USS and HMS can only means the origin of the buidling of the ships. Klingon ships, at least in the Star Trek Online game, get IKS (Imperial Klingon Ship) before its name.

NCC doesn't stand for anything. It was devised by Matt Jefferies, art director of the first Star Trek series. Jefferies, who is a pilot, based NCC on 20th century aircraft registration codes. In such 20th century usage, an "N" first letter refers to an aircraft registered in the USA. A "C" second letter refers to a civil aircraft. Jefferies added a second "C", just because he thought it looked better. Think of it as being like the arbitrary three-letter code that's part of automobile license plate numbers in many states. Also NX means "Naval Experimental".

In one of the episodes of the original Star Trek,Mr.Scott refers to the Enterprise as "a million gross tons of vessel".Can't recall which episode though.I've been curious myself as to how the star ship would compare in size to the aircraft carrier of the same name.Saw the Enterprise aircraft carrier once in Seattle.Big but not as big as I imagined.
