Yes it is, in the Dark Tower it's said that all the worlds exist on different levels of the Dark Tower, Desperation takes place on one, The Regulators on another. There's some references to the Crimson King, the Dark Tower's villain, in Desperation, as well as connections to King staple, Flagg.

I'd just like to add that there's more than just those two novels that are connected to the DT series. 'Salem's Lot, The Stand, The Talisman, IT, Insomnia, Black House, Hearts In Atlantis, and Everything's Eventual to name a few, whether it be a person or place or a thing. Especially Insomnia, which talks a lot about the Crimson King, and Hearts in Atlantis, and 'Salem's Lot. Those three are very closely connected to the DT series. I would suggest you read them before starting the series.
