Assuming you are coming from Ho Chi Minh city (which is the route most people take), it's about 2 - 3 hours to the border. From there, you take the bus to Phnom Penn, another 6 or so hours. Of course, this is all dependent on how much traffic there is, and on ferry running times once in Cambodia. (some rivers dont yet have bridges)
You can easily do the trip in one day. A word of advice: Do not attempt to stay in the border town on the Cambodian side of the border. It's a dump, very limited in accomodation, and full of dodgy casions. Just go straight through to PP.
