Depends on the company. For example, Coca-Cola ONLY responds to applications via Internet. They even only consider present employees for transfers via Internet applications. (I have a good friend that works for Coca-Cola.)
Other, smaller companies, may want you to come in person. Normally, the help wanted ad says how you should apply - in person, faxing a resume, sending one to a mailing address, or on their website. I know one thing. I have never once got a response out of Career Builder, Monster, etc., so don't waste your time posting resumes on such sites.

Applying in person. You can express ethusiasm, being professional, and show respect through body languge. In addition, you can get a good or bad vibe by dealing with a person or a comany while in actual presents. This is basedoff of my own personal situations.

It's best when you can find the position before it is posted on the internet.
Why?
Less competition. If you're qualified for the position, you'll save an interviewer time and hassle.

I say over the Internet. They will call you if they are interested in hiring you. if you know of any companies that are not hiring, then perform an informational interview. that is how you do it in person.
