Well it really all depends what you are looking for.
Sanibel Island is really great if you want peace and quite, Nice white beaches not a lot of people. Good places to walk. The Lighthouse Cafe has the best breakfast and the Bubble Room is a fun place for dinner. You will also find the best shelling in Florida here. Fort Meyers is near by which is good for shopping and getting deep sea fishing tours. The island is very small has some really neat local shops.
St. Augustine: Good place for families or young people. There is a lot to do. The old fort is very cool, surfing lessons for the Pit surf shop great! Good food at Scarlet O'Hara's. They also have some really good tour companies that do great city tours or ghost tours. St. Augustine is cool because it is the oldest city in America. There is always some kind of re-enactment going on. The change of the Guard is very neat to see. There is also Tavern De Gallo which is the oldest bar around, they have a group of guys that sing sea shanties on Friday and Sat nights very fun. Pirate Haus great place to stay downtown free parking cheap rates. Also St. George street good fun check out the St. George Tavern for good drinks! They also have the Alligator farm which is just too cool. The lighthouse is very neat and the people that work there know there stuff. Plus you are 45 min away from Daytona and 2 hours away from Kennedy. Nice location. The first Ripley's Believe it or Not is here. Yeah it is a little touristy but still very cool to see.
Of course Orlando if you like the theme parks is always fun. Haven't really done much else there.
Miami, lived there fun city, depends on your age it also depends on what you are looking for. If you are going to party this is the place to go. Kinda expensive and sometimes overated.

Orlando if you like theme parks
Panama City if you are younger and like the party atmosphere
Destin if you like crowds, good places to eat and nice beaches...family activites
St George Island or Cape San Blas if you like peace and quiet and like being off the beaten track...nice beaches and nice houses to rent.
Navarre Beach...beautiful, close to shopping and right on the beach
Okaloosa Island...less crowded than Destin, nice beaches, close to several good places to eat.
Wakulla Springs if you REALLY want to experience old FL...1930s hotel, no TV, springs to swim in complete with alligators...(located near Tallahassee)
I personally like all the above places and have enjoyed them all more than once.
