The length of light waves absorbed by a substance. In a nut shell, the machine shines light through a sample of a compound; you set the wavelength, and then the machine measures how much was absorbed by the sample.

It's useful for comparing the observed data of a substance to known data, giving a sense for the purity of something, or how concentrated the compound of interest is. This can also give you a precise readout of what color a compound is (say an ink or dye).

The top two are correct, but I think what you are asking is what specifically will it measure. The answer is: with many different assay kits, you can measure protein, cDNA, mRNA, and triglycerides. There are probably more things you can measure with this, but these are probably the four most commen.
