You would use a combination of two methods. First, use a magnet and see if the nail responds to the field. If the nail responds, you know it contains some kind of ferrous compound.

Once you determine the nail is magnetic, the next step is to scuff it up a bit with some steel wool. If the scuff mark remains black, the nail is iron. If the scuff mark is bright and shiny gray, the nail is steel or some other iron-containing alloy.
