I think she suspects but she doesn't know for sure. Like in the second movie when Peter decides not to be Spiderman and she tells him that Pablo wonders where Spiderman is and that the world needs heros. So, maybe she suspects or maybe she said it because Peter takes pics of Spidey and she hoped he would pass the message along. Who knows?
