You can have the test before the baby is born but why risk it ?
Just wait till the baby is born. Tell your doctor in advance you'd like this test done.
You will also need a blood test from the men involved.
Goodness.

It is possible to do it before the birth, but safer to wait until the child is born.
After birth, a test can be performed immediately - it is fast and painless. They can take either a pinprick blood sample from the child's heel or a mouth swab.
As far as consent is concerned, this is a legal matter that varies around the world. You may need the consent of the mother to do the test, you may be able to have it performed without. However, bear in mind that DNA testing is usually more straightforward and more accurate if you have DNA from both parents and the child than just the father to test against. Single parent testing is normally the exception rather than the rule - as well as the legal implications of this, many labs also have a code of ethics to work under and wil not test a father and child sample if the mother is not involved/consenting.

can be done at birth....as far as the procedure legally it depends on the laws where you live.....the mother will have to sign a consent, if shes not willing to do so your brother will need a court order more than likely. the problem with that is until he can show that she put his name on the birth certificate or she asks for child support probably no judge will order it. however if she does indeed put his name on it he can wait till after the baby leaves the hospital and take the baby and the birth certificate in and have it done without her consent. like i said though that is all dependent on the laws where hes at.

If she is still pregnant there are alot of risks getting the dna test now, but if she has already had the baby, then she or they can get it done asap. Also if she is pregnant now, then you can wait until she delivers and they can take dna from the baby's imbelical cord when it is born along with fathers dna to determine if he is the father or not.

They can test the tissue of a fetus who is aborted.
A live birth child can be tested immediately. All they'll need is a swab from the inside of his cheek.
