65 teams. There is an opening round game for the "traditional" 64th spot in the tournament. The game was started in 2001 when the Mountain West Conference had split from the Western Athletic Conference in 1999. Under NCAA rules a new conference has to serve a 2 year probationary period before receiving an automatic bid to the NCAA tournament. Once the Mountain West received their automatic bid the field was increased to 65.
Even though people refer to the game as the "play-in" game, it is not officially referred to as the "play-in" game because the teams are in fact tournament members before the game is even played.
Due to the fact that most people still consider the tournament to be a 64 team set, a lot of people don't even recognize the "play-in" game on bracket picks.

there are 65 teams. there are 4 different sections in the tournament, each with a 1-16 seed. and there is one game before march madness with 2 16 seeds that play for the final spot in the march madness tournament. the winner of each section goes on to the final four. the winner of the remainind 4 is crowned the champion.

65, the first game played is a play in game at which point the winner is classified as a number 16 seed and plays the number one overall seed which this year i believe plays in the midwest bracket.
