I am trying to caculate how much sodium hydroxide 50% (by wt) to a solution of 500L of water and 60L of ACN to get a final pH of 10. ACN is soluble with Water but will it drop out when the NaOH is added?

Depends on how much ACN you add. If you use ~50% or less of ACN then it won't really matter, the NaOH will dissolve only in the water and essentially the ACN will not affect the pH much. So ignore the ACN except in the sense that it reduces the actual volume of the water (i.e. 1M NaOH in water = 2M NaOH in the water part of 1:1 water:ACN if you add the same amount of NaOH)
