You cant, to the extent of my understanding, merge two physical Disks. You can join two partitions however. Using a program like:
- Partition Magic (not free)
- Acronis Disk Director (30 day free trial no limitations)

If you would like see only the C-drive instead of two that you may achieve this by creating an empty directory then go the your computer management console (right click my computer->select "manage"->disk management), right click on the disk and when going through the formatting options, you will be have an option to mount the disk to an folder.

I agree with gerosna's answer. But if your computer support RAID 0 or disk striping then it is possible to do that. Some personal computer have a special card which support RAID, although this card ussually reside in server system.
