Killer question. Lesbians don't come in just one way, they're all different. :P I mean, take a prudish, matronly woman. Gay? Yes, yes she is. A tomboy, sex kitten, schoolgirl, etc.
You cannot tell someone's sexuality by looks alone, it is definitely more than that.
Talk about it, ask other people (don't believe everything you hear though, but she may be open with her sexuality). Hit on her a little, see if she does the same back. It's... body language.

When people find out I prefer women they always say "You !! No way!!" Cause I'm so femme, I love feminine women and I just approach a hot girl and keep my fingers crossed :) I can tell they are bi or lesbian if they give me their number :o)

U wont know un less she till's u cause u cant just look at someone and know there sexual oration but some times u can till there straight by if they only dates the opposite sex but they might actual be bi instead and decided to not date the same sex.
