There are two routes to drive from Gurgaon (Haryana) to Jodhpur (Rajsthan). The first one below is just 30 kilometers longer than the second one, but it is faster and easier (Better highway and speed). I would suggest you take only the first route.

I would take the second route ONLY IF I were interested in the interiors of Rajsthan and watching the rural rajasthan closely.

First Route: (Approx 580 Km):

Take NH8 until Beawar (You'll pass Jaipur and Ajmer). Gurgaon to Beawar is about 420 Km.
From Beawar, turn right on NH14.
After going about 22-23 Km, again turn right on "State Highway (SH) 5" towards Jodhpur (distance is about 130 Km).

[NH8: Delhi - Jaipur - Ajmer - Udaipur - Ahmedabad - Vadodra - Mumbai Highway; NH14:Beawar - Sirohi - Radhapur Highway]

Second Route (About 550 Km):

Take NH8 until Kotputli (about 125 Km from Gurgaon)
At Kotputli turn right and go about 45 Km and then turn left on SH13 (go 15-16 Km)
At SH 13 Govindpura, Turn right - go 8-9 Km
At Dehrajori Wali, Turn left - go 5-6 Km
At Kotri Luharwas, Turn right - go 10-12 Km
Turn left on SH 20 - go 35-40 Km
At Sikar, Turn slightly left - go 30-32 Km
At Losal, Turn right - go 17-20 Km
Turn left on SH 18 - go about 80 Km
Turn right on SH 2 - go about 40 Km
Turn slightly left on NH 65 - go about 120Km and reach Jodhpur.

So, now you see why I suggest taking the first route. Even the long-distance buses take the first route. I just wanted to mention the second route for your information. Second route saves you 30-40 km but it'll take you longer time to drive to Jodhpur.
STICK TO ROUTE ONE and you'll be driving mostly on NH8 (which id good) for about 420 Km and only about 160 Km on SH 5.

From Gurgaon take NH 8 - Delhi to Mumbai Road.

The route would be -
Gurgaon - Kurkola - Daruhera - Kotputli - Shahpura - Jaipur - Mahlan - Kishangarh - Ajmer.
From Ajmer take right to Jodhpur.
