Get bit by a radioactive spider, obviously. Make sure it's blue and red though.
Other things you can try...
buy yourself a magic sword like He-Man's;
have your midichlorine levels checked to see if you can train to be a Jedi;
get really close to an exploding comet;
Get struck by lightning (warning: not recommend because this resulted in Powder turning into a lightning bolt at the end of the movie);
You can drink some magic potion for a Jekkyl and Hyde experience;
You can inhale the dust cloud from a nuclear explosion.
Oh yeah, and apparently a brain tumor will do it (not sure of where to get one though).
