When I worked there many, many years ago stocking was done after the store closed in a fun time they called 'zoning'. Each store area's assigned employees determined what was needed, brought out the available merchandise, then re-stocked the items on to the displays. Additionally, the remaining items after re-stocking was complete were then 'fronted' to fill in all the gaps on the shelf or rack. Yeah, I worked in Health and Beauty...all those tiny medicine bottles and odd shaped gifts....man oh man was that a Summer to remember.
Any super fast moving items (like notebook paper during the back to school sales) were re-stocked during the day as well if they became depleted.

It depends on which section of the store you're talking about and during which time of the year. Books/DVDs/CDs get stocked before opening on Tuesdays (when new DVDs/CDs are released), most of the year everything else gets stocked before or after closing and during the holiday season, most things are re-stocked throughout the day and they have people work all night to re-stock before opening the next day.
