The link below will help answer your question. Its all about how to drill holes in mirrors. Your definately going to need a drill bit specifically designed to drill through. they are generally diamond dust tiped bits. And never apply to much pressure while drilling glass. Let the weight of the drill do they drilling. Good Luck with your project, its very tricky.

Drilling a mirror is relatively easy to do if you take care and plan ahead.
First use only a HHS metal drill bit, do not try it with a masonry bit.
Second use masking tape and cover the place where you want to drill the hole.
Third support the back of the mirror ideally with thick cardboard, this will prevent the glass from flexing and breaking.
Fourth have the drill on the fastest speed possible and don't press hard on the drill.

Lay the mirror on an old piece of carpet. Put two strips of decorators masking tape in a cross on the center and mark where you want to drill. Use a small wood drill in a cordless drill and gently start drilling without hardly any pressure. Once you have gone through the tape you should have made a slight score on the mirror, then switch to the smallest masonary drill you can find and using plenty of speed but very little pressure start drilling. It will take you a while but it will work.
Alternatively, take the mirror to your local glazers and pay them to do it.

At your local hardware store they should have a glass bit. Or better yet if you have a glass store in your area they may be able to get you one. There similar to an oval end that comes to a point and there coated with diamond chips, very small. There not very expensive but they are what you need to drill this hole. The bit is made expecially for drilling glass.

You need a drill that is called a"spade drill", made of HSS (high speed steel) it looks sorta flat on both sides kinda like a flat screw driver ecept with flat points on the other side, then put a dam of clay around where you want the hole and use terpentine as a coolant, use about a 2 or 3 hundred rpm for the drill and you'll be in business..I've done this for years ......

You can purchase glass drillbits at larger hardware stores. I make a ring out of putty around the area I will be drilling in. Pour in a small amount of isopropl alcohol for cooling the bit. Drill with light pressure. I have also added a small amount of salt in the alcohol for more abrasion.

You can't do it with shatteing the mirrow. Find a commercial glass shop and ask if they can do it. They use high speed diamond tip drills with a cooling liquid to cut thru the glass/mirror.
