You may want to try doing webcomics/self-publishing to get feeback and maybe buildup a reputation and fanbase. I am not really sure what the demand for American manga artist is, but I believe a popular American one is Adam Warren. You may want to go to college to learn how to properly get your ideas down on paper, which is what I am doing next year when I study illustration (in hopes of drawing traditional American comics). I think your best bet is to try and come up with something that is both visually unique as well as a great story so that it does not blend in with the generic anime look.

I would say the best place to try is at Comic Book Conventions. Publishers set time aside to look at peoples submissions and even critique them. You will have to stand in long lines but it can be worth it to reach your goal. Try it at big ones like the San Diego Comic Con. http://www.comic-con.org/
