By far the first has to be Jimi Hendrix. Everyone had him on their list. One thing that dissappointed me was that no one has John Frusciante on their lists. He is regarded by many as the best guitarist in business and his riffs are near impossible to learn. I know he hasn't been around for forever, but the RHCPs have been around for quite a while.

It depends what music you like, my preference for popular music is Eric Clapton. However, there are many other great guitarists.
Classical: Yorgos Foudoulis
Electric Jazz: Les Paul
Accoustic: John Williams

this is one of those imposible questions where everybody votes for their favourite regardless of whether they are actually the best. so heres my tuppence worth.
my favourite is pete townshend, however i think the best guitarists are:
django rienhardt for his stunning technique
jimi hendrix for completely taking over the instrument
eddie van halen for bringing the guitar into the eighties
i think these three have been the most influential in regard to how they've moved the instrument forward, i'm not neccesarilly a fan, i just respect and understand their contribution.
