Descartes uses sytematic doubt to discount knowledge that he can not be sure of. If there is a real possibility for doubt about something known then he will discard that thought for the time being and search for something more fundamental of which he may be certain. He believed that if we start with doubtable knowledge then all subsequent knowledge would also be suspect to doubt as well. In short, he used doubt as a test to search for certain knowledge.

Decartes began by eliminating everything around him that may desive or trick him. Or all things that he has doubt about. Till he got down to the core the one absolute thing that could not ever be doubted. Then he felt that is ultimate knowledge. From that he would again build from that point back up. This he thought would mean that all knowledge after that would be undoubtable. I personally think he failed. But he did come down to, "I think therefore I am."
