These days their can be so much competition for the good jobs that your cover letter needs to be great otherwise they won't even bother looking at your CV - but I'm guessing you know that by your question.
I'm applying for jobs right now and what I've found is that you should
a) Begin by stating what job you are applying for (is their a reference number, date posted, what webiste/paper etc) and briefly why you want the job or what attracted you to the job.
b) Your CV goes into a lot more detail, however you need to link your CV to the skills and experiences needed as per the job description in the middle paragaph(s) of the cover letter.
Just pick out a few points that highlight why you are what they are looking for, you need to back this up with actual evidence/examples to prove this - be specific but brief.
Very Basic Example:
I gained broad exposure to a number of [skill you have that the company in question are looking for] at [company/uni] as a [previous job title]. My responsibilities ranged from developing [something you did that was listed in the job description], maintaining [another experience or responsibility that is relevant], which as I understand would be required in this position.
c) To finish off the letter: state your CV is attached, request interview or perhaps mention you will call/email in x days. Basically just rounding it off nicely.
My example (by no means perfect!):
I welcome the opportunity to discuss my suitability further at an interview, please find my CV attached. You can reach me on xxxxx; I look forward to your response in due course.
d) They can be really hard to write, but you can get some help out their if you search. The only problem for them to be relevant it needs to be specific to you and the job you apply for, I got some examples off the net, listed my skills and experiences then picked thorugh the job description for things that matched up.

It is called a "cover letter" and it all depends on what sort of conveyance your letter will be covering.
Usually, a cover letter is for a resume.
You can find plenty of guides on the internet on how to write a cover letter and all kinds of other business correspondence.

To be truthful there shouldnt be that much in a covering letter just saying where you read the advertisement, that you are interested and please find enclosed a copy of my CV. you should never go into detail with a covering letter, its the CV you put in with the covering letter that is important

A covering letter for what? If you mean a CV then you simply write to advise that you're attaching your CV regarding blah blah job and that you hope to hear from the soon.
