I'm writing a letter to a company. I would like to ask you which type of letters should I use in the Subject field, eg.

Subject: Application For The Position Of

or

Subject: Application for the position of

I think either is fine, although I usually use Title structure (capitals at the beginning of each word) when writing a formal e-mail.

it sounds more professional

Don't write the whole message in the subject field.. Because if the job has been taken, nobody will bother to open your message.
Just write: Job Application

This way you make people read it even if the position you're applying for had already been taken.

Formal letters do not have a "subject." Memos do. In which case, the use of mixed case (the second one) is more correct.

Ending in "of" is also not correct. I presume that you meant that an actual position (e.g., "application for the position of clerk") would be there.
