You're best bet is to use the tools already available to you in your operating system. If you're a Windows user, you can do the following:
Make sure that you have a password set for your username on your computer. If you don't have one set, go to Control Panel -> User Accounts and follow the directions to set a password.
When you're using your computer and want to lock it when you get up to leave, you can log out or just hit the Windows key (between Alt and Ctrl) and the letter L at the same time. This will lock your desktop until you return and type in the password.
You can also set it to lock automatically after a period of time you define in the case that you walk away and forget to lock it. It's an option that's part of the screen-saver settings. Right-click on your desktop to bring up a list of tasks and options. Click on "Properties". In the window that pops up, click on the screen saver tab. In that tab, check the box that says "On resume, password protect" and set the screensaver to something other than "none". You'll want to set the wait time to be pretty low, i bet, to minimize the chances of someone walking by and having a go.
If you're using a Mac, I'm sure there are options to do that, i don't have a mac in front of me to give you step-by-step instructions.

Microsoft,shockingly, was one step ahead of you. This feature is already built into XP. Just right click on your desktop and click properties,click screensaver tab and check on resume, password protect. May be different depending on your OS. This is based on Window XP Professional. Hope this helps
