if the culture is obtained from a body surface ( inside or outside), human cells may be included in the culture. are human cells gram-positive or gram-negative?

Gram negative. Only cells with a cell wall are gram-positive.

The Gram stain reacts with the cell walls of certain bacteria and dyes them bright purple. Human cells don't have cell walls. Therefore when human cells are exposed to Gram stain there isn't any color development. So human cells would be classified as gram-negative.
