It is areference to Opera. Often times in days past female operatic singers were large women. Many operas ended with a female with a particularly powerful voice (usualy a large woman and they did tend to have very powerful voices) singing the finaly, hence the phrase "the opera isn't over till the fat lady sings."

The reference is from Kate Smith a popular entertainer from the 50's would sing the national anthem after a victory in Philadelphia at the National flyer's hockey team arena.(during the 70's] she was a stout woman hence the term" fat lady sings.'

It is a term from opera. the "fat lady" is because many historical ( and even current) opera figures were heavy men and women. so if the show isn't over til the fat lady sings it is a reference to the fat woman's closing song.
