Falco: Complete the 100 man melee
Marth: Use all 14 original characters to complete Classic or Adventure
Roy: Complete classic or Adventure with Marth
Ganondorf: Win event match 29 (Trifroce Gathering) (I'm not 100% sure of the match number. Go by the name)
Pichu: Beat event match 37 (Legendary Pokémon)
Mewtwo: Play multiplayer for a total of 20 human hours (Set up a four player match with no CPU and leave it running for 5 hours)
Mr.Game and Watch: Once you've unlocked the other characters, beat Classic or adventure with all of them.

Bascially..all you have to do is beat classic with every character.
To get Mewtwo...the easiest way is to get 4 controllers and choose Link for all of them...make the melee timed and choose Hyrule Temple...then leave the game for a couple hours and when the game is over..Mewtwo will challenge you..
Or you can play 700 v.s. battles >_<
Everyone else is so easy to get...

There is very many ways to unlock characters.
one way is keep playing vs battles
another way is to beat the classic and adventure mode with everyone, beat All-star mode

the answer is not the simple one you want. you must play, play, play. do everything with every chacter. that is the ONLY way to unlock everyone, including mr. game&watch.
