Well, when you squeeze the tube, the pressure on the other end causes the toothpaste to go up the tube.

The tube of fixed volume is filled with a liquid. When you squeeze the tube, you effectively change the volume, forcing the toothpaste through the top.

Notice that if you squeeze a full tube with the cap on, nothing happens. You are trying to change the volume of the tube by squeezing it, but since the cap is on, the toothpaste won't squirt out. This is when you're effectively pressurizing the toothpaste. And if you squeeze really hard with the cap on, you will eventually overcome the strength of the tube, and it will burst.

The paste is a liquid and tries to retain a certain volume. Meanwhile since it is a liquid it can easily change its shape. When you squeeze the tube, you cause the volume of the tube to decrease. the paste takes the easiest path to retaining its volume and comes out through the nearest opening. Which is also why when an older tube cracks, the paste will squirt out through the crack much faster than from the opening.

The explaination is the law of Pascal - the pressure of a fluid to the walls of the vessel are equal in all directions. So, there is only one hole, though which the fluid can go out. And it does that..
